Geboren en getogen in Kent in het zuidoosten van Engeland, volgde Arterton de Royal Academy of Dramatic Art (RADA) in Londen. Ze speelde in een aantal theater stukken aan de RADA en heeft daar ook haar podium debuut gemaakt.
Haar filmdebuut was in de recent uitgebrachte film st. Trinian's, geregiseerd door Oliver Parker en Barnaby Thompson. Arterton speelt Kelly (The Head Girl) in deze moderne versie van de originele St. Trinian's film. In de film spelen verder een aantal bekende Britse sterren, zoals Rupert Everett, Russell Brand en Colin Firth.
Vlak voordaat ze aan Quantum of Solace begon, was Arterton net klaar met het draaien van 3 And Out, geregiseerd door Jonathon Girshfield. Arterton speelt de rol van Frankie in deze komedie, naast Imelda Staunton en Mackenzie Crook. Verder speelde ze vorig jaar ook in de Engelse misdaadfilm Rocknrolla, geriseerd door Guy Richie, met mede hoofdrolspelers Gerard Butler, Thandie Newton en Mark Strong.
Op televisie, is Arterton het laatst te zien geweest in Stephen Poliakoff's Capturing Mary (BBC). Ze is ook net klaar met de opnames van Lost in Austin voor ITV, geregiseerd door Dan Zeff, waar ze de rol van Elizabeth Bennett speelt naast Jemima Rooper, Alex Kingston en Tom Riley.
Op het podium was zij voor het laatst te zien als Rosaline in Love's Labour's Lost, geregiseerd door Dominic Dromgoole in het Shakespeare's Globe Theatre.
